It cleaves the phosphodiester bond between the 5′-ribose of a nucleotide and the phosphate group attached to the 3′-ribose of an adjacent pyrimidine nucleotide. The resulting 2′, 3′-cyclic phosphate is hydrolyzed to the corresponding 3′-nucleoside phosphate. Unhybridized RNA regions in DNA:RNA or RNA:RNA hybrids can be removed; It can be used to determine the location of a single-base mutation in DNA or RNA, annealing the labeled RNA probe with the DNA or RNA containing the single-base substitution to be examined, and the mismatched bases in the RNA:RNA or RNA:DNA can be cleaved by RNase A.
