Introduction
This kit uses a pair of primers, Taq DNA polymerase, four monomeric nucleotides (dNTPs) and other components to amplified the CHD gene of the bird sex chromosome by PCR technology, and the sex of birds is identified by agarose gel electrophoresis.
This kit is only suitable for the sex identification of non-flat-chested birds, most of the existing birds belong to non-flat-chested birds, and flat-chested birds only include ostriches, emus, cranes, ostriches (American ostriches), and kiwis.
Precautions
In order to ensure the accuracy of the experimental results and avoid contamination, it is recommended to set the following four experimental areas from the preparation of the reaction solution to the addition of the test samples and physically extract them. In the 1-3 area, it is strictly forbidden to open the PCR tube containing the amplification product.
Area 1: Preparation and dispensing of PCR solutions (positive pressure requirements in the laboratory).
Area 2: Preparation of nucleic acids for the test sample (negative pressure requirements in the laboratory).
Area 3: Add the test sample to the reaction solution and perform the PCR (negative pressure requirements in the laboratory).
Area 4: Electrophoretic detection (! The laboratory has negative pressure requirements).

Result: according to the Figure 1, the sex of these bird samples(1,6,11,14,15,21) are female, the other samples are male.
