实测simgen粪便DNA保存液的保存时效

粪便DNA保存液作为Simgen的热销产品深受用户喜爱,咨询的客户也很多,之前就有客户问到产品介绍中描述的“
粪便样本中的DNA经粪便保存液稳定后可在室温(15-25℃)保存6个月以上而免于降解
”有没有具体的数据资料。当时分光光度计的检测数据有记录,但是电泳图并没有保存,而且当时取样量不多,只做了26周测试,后面就因为样本不够而无法继续进行下去了。为了提供完整可信的实验数据以及探究粪便DNA能不能保存更久的时间,小新采集了样本进行了粪便DNA保存液保存效果的长期测试。
实验目的:
测试粪便DNA在Simgen粪便DNA保存液中长时间保存的效果。
实验材料:
新鲜采集的人粪便、粪便DNA保存液(Simgen Cat.No.4103050)、粪便DNA纯化试剂盒(配套粪便DNA保存液)(Simgen Cat.No.4111050)超微量电子天平(DENVER INSTRUMENT,TP-213)旋涡振荡器(越新仪器,XH-C)电热恒温培养箱(上虞市沪越仪器设备厂,DHG 303-0)电子恒温不锈钢水浴锅(上海宜昌仪器纱筛厂)台式离心机(eppendorf Centrifuge 5415 D)超微量分光光度计(Simgen Cat.No.sim100)电泳仪(北京六一仪器厂,DYY-6C型 )
实验方法:
小新于2022年9月19日采集了5 g粪便样本,加入了10 ml粪便DNA保存液,旋涡振荡直至粪便颗粒全部分散溶解后,置于37℃恒温箱保存至今(除了过年和十一长假,其他时间恒温箱一直开启),期间每隔一段时间提取一次DNA。 DNA提取操作步骤如下:1. 将吸头尖端剪掉,从粪便DNA保存液保存的粪便样本中吸取100 μl悬浊液,转移到自备的1.5 ml离心管中。2. 加入350 μl 70℃温育的Buffer HST,盖上管盖,混合均匀,70℃水浴5分钟。3. 12000 rpm离心1分钟。4. 吸取20 μl蛋白酶K贮存液,加入到一个1.5 ml离心管管底。5. 吸取200 μl步骤3中的离心上清加入到该1.5 ml离心管中。6. 加入200 μl Buffer SL,旋涡振荡约15秒混匀。将离心管置于70℃水浴10分钟。7. 加入200 μl无水乙醇,温和地翻转4~6次混和均匀。低速离心数秒使管盖上的溶液沉降到管底。8. 吸取步骤7中的混合液加入到核酸纯化柱中(核酸纯化柱置于2 ml离心管中),盖上管盖,离心弃滤液。9. 依次用500 μl Buffer WA和600 μl Buffer WB洗涤纯化柱。10. 高速空离去除残留液体。11. 弃2 ml离心管,将核酸纯化柱置于一个洁净的1.5 ml离心管中,在纯化柱的膜中央加入150 μl 70℃温育的Buffer TE,盖上管盖,室温静置1分钟,离心得到DNA。
实验结果:
1、在超微量分光光度计上用试剂盒内的Buffer TE调零,测量洗脱下来的DNA,结果如下:

分析与讨论:
1. 粪便样本经粪便DNA保存液处理后,置于37℃保存至今已达16个月,从分光光度计测量结果来看,每次均成功提取到了DNA,不过浓度略有跳动,总体趋势上看似乎放得越久浓度越高。2. 经过电泳检测分析,当天提取的粪便DNA浓度偏高,应该是由于样本刚采集,提取到的粪便DNA中混有一些降解的RNA提高了OD
260
的吸收值,这与OD
260
/OD
280
的比值接近2.0也有对应关系。3. DNA提取的浓度随粪便样本室温放置的时间延长浓度有升高现象,这是出乎意料的结论。经过认真分析我们认为是以下三个原因所导致:1) 粪便DNA保存液保存粪便DNA的过程中在缓慢地溶解细菌,随着时间的延长,释放到保存液中的DNA就越多,在后续提取DNA的离心步骤(步骤3)中被沉淀丢失的DNA就会越少,显示出DNA的回收量就越高。2) 在37℃存放的条件下,释放到粪便DNA保存液中的DNA还是会缓慢地分解的:这个从电泳分析图中就可以观察到,存放1~4个月的样本中提取的DNA拖尾做少,在存放5~16个月的样本中提取的DNA中,比较明显的拖尾就观察到了。DNA的片段越小,相对而言OD260的吸收值就越高,造成了提取的DNA浓度偏高的假象。3) 最后我们发现,随着37℃放置时间的延长,水分会有部分被蒸发,这就相当于粪便DNA保存液被浓缩了,这很可能是16个月存放的样本提取到的DNA浓度有明显提高的主要原因。4. 好消息就是:经Simgen粪便DNA保存液保存的粪便,在为期16个月的测试中,粪便DNA虽然产生了一些拖尾,但是基因组DNA的主条带依旧清晰可见,无需担心后续的分子生物学实验应用。综上所述,我们可以得出结论,粪便样本中的DNA经Simgen粪便DNA保存液稳定后至少可在37℃保存16个月而免于降解。如果将该样本存放到2~8℃,毫无疑问疑问保存时间可以延长到2~3年。至于加了Simgen粪便DNA保存液的粪便样本在常温是否还能保存更久,小新会将测试继续下去,敬请期待。
